چکیده

Several products of CTL and NK cells, including a pore-forming protein (PFP;t perforin or cytolysin) (1-4) and several serine esterases (SE) (5-12), have been cloned and sequenced . These proteins are localized to cytoplasmic granules and represent the known structural components ofthe pore-formation and granule exocytosis models for cell-mediated killing (13-17). Granule mediators are expressed by CTL and NK cell lines maintained with IL-2, but there is still little information on primary effector lymphocyte populations. Moreover, to date, most ofthe studies have been performed in the murine system . Here, using cDNA probes encoding two SE (SE 1/granzyme A/Hanukah Factor and SE 2/granzyme B) and the recently cloned perforin cDNA, we have initiated studies aimed at understanding how the production of these proteins is regulated in human primary T lymphocytes. We have examined RNA accumulation in response to a number of well-known T cell mitogens and have determined the effect of Cyclosporin A (CsA) on this inducible response.

Serine esterase and hemolytic activity in human cloned cytotoxic T lymphocytes

Target cell lysis by most murine cytotoxic T lymphocytes appears to be mediated by a complement (C9)-like protein called perforin, contained in high-density cytoplasmic granules. These granules also contain high levels of serine esterase activity, which may also play a role in cytolysis. Analysis of 17 cloned human cytotoxic T lymphocytes revealed the presence of serine esterase that is very si...

Resistance of cytolytic lymphocytes to perforin-mediated killing. Lack of correlation with complement-associated homologous species restriction

CTL and NK cells cultured in vitro are known to produce a cytolytic pore-forming protein (PFP, perforin) localized in their cytoplasmic granules. Using purified perforin, we showed here that both cloned CTL and primary killer cell populations, including allospecific CTL, NK/lymphokine-activated killer cells, and MHC-non-restricted CTL, were more resistant to perforin-mediated killing than other...

Expression of perforin and serine esterases by human gamma/delta T cells

gamma/delta T cells have recently been described in association with a number of disorders, including autoimmune diseases. gamma/delta T cells are thought to play a cytotoxic role, but their mechanism of action is not known. Several granule mediators of cytotoxicity, including a pore-forming protein (perforin), and a family of serine esterases, have been isolated from cytotoxic T lymphocytes (C...

Phosphatidylinositol 3-kinase-dependent and -independent cytolytic effector functions.

Two distinct forms of short-term cytolysis have been described for CD8+ CTLs, the perforin/granzyme- and Fas ligand/Fas (CD95 ligand (CD95L)/CD95)-mediated pathways. However, the difference in signal transduction events leading to these cytolytic mechanisms remains unclear. We used wortmannin, an irreversible antagonist of phosphatidylinositol 3-kinase (PI3-K) activity, to investigate the role ...

Induction of target cell DNA release by the cytotoxic T lymphocyte granule protease granzyme A

The rapid breakdown of target cell DNA during CTL-mediated lysis has been difficult to explain by the granule exocytosis model of cytotoxicity. The involvement of CTL granule proteases in this process was strongly suggested by experiments in which CTL were pretreated with the serine protease inhibitor PMSF, in combination with agents that raise the pH of acidic intracellular compartments. While...
